MMPOWER-3: a larger trial that did not meet its primary endpoints

Negative primary results are an important part of an evidence library.

01 / DESIGN

What was studied?

MMPOWER-3 randomized 218 participants with genetically confirmed primary mitochondrial myopathy to elamipretide or placebo for 24 weeks.

02 / FINDINGS

What did the researchers report?

The study did not show an improvement over placebo in its primary endpoints: six-minute walking distance and total fatigue score. Most adverse events were mild or moderate.

03 / INTERPRETATION

What are the limits?

Tolerability is not the same as demonstrated efficacy. These findings concern this disease population and study design; they do not establish benefits for healthy aging, athletic performance or other untested uses.
