Early MMPOWER findings: why the statistical analysis matters

A small early study illustrates the difference between promising signals and confirmed outcomes.

01 / DESIGN

What was studied?

This phase 1/2 study randomized 36 adults with genetically confirmed primary mitochondrial myopathy and assessed walking performance after five days.

02 / FINDINGS

What did the researchers report?

The highest-exposure group showed a larger walking-distance change, but the unadjusted comparison with placebo had a P value of 0.053. Other analyses suggested a signal that warranted further investigation.

03 / INTERPRETATION

What are the limits?

The short duration, small sample and differing analyses require caution. The later MMPOWER-3 trial is essential context; a positive exploratory signal should not be presented as established clinical efficacy.
