Tesamorelin continuation and withdrawal: interpreting a blinded extension

A second trial used a rerandomized extension to investigate what happened after initial treatment.

01 / DESIGN

What was studied?

This 12-month study enrolled 404 people with HIV and excess abdominal fat. After a six-month randomized phase, participants receiving tesamorelin were rerandomized to continue or change to placebo, while the initial placebo group changed treatment.

02 / FINDINGS

What did the researchers report?

The initial comparison showed reduced visceral fat with tesamorelin. The extension examined maintenance and reversal of that response, alongside body image, IGF-I, glucose and safety measurements.

03 / INTERPRETATION

What are the limits?

Extension results answer a continuation question and should not be presented as a permanent change after stopping. This publication reports a particular phase 3 trial, distinct from the earlier 412-participant study. Neither trial establishes equivalence of laboratory catalog vials to the pharmaceutical study product.
